胆固醇酯酶催化胆固醇酯水解生成游离胆固醇（FC）和游离脂肪酸（FFA），胆固醇氧化酶催化游离胆固醇氧化生成∆4-胆甾烯酮和H₂O₂，过氧化物酶催化H₂O₂氧化4-氨基安替比林和苯胺类似物生成紫色化合物，产物在550 nm处具有特征吸收峰，通过吸光值变化即可定量检测总胆固醇的含量。
