亚硝酸还原酶能够将NO₂^-还原为NO，使样品中参与重氮化反应生成紫红色化合物的NO₂^-减少，重氮化反应产物在540 nm处具有特征吸收峰，通过吸光值变化即可表征土壤亚硝酸还原酶的活性。

①若测定吸光值超出标准线性吸光值范围：高于最高值建议适当增加样本量或将离心上清液适当稀释后再进行测定，低于最低值建议适当减少样本量后再进行测定，计算时相应修改；
