结合态淀粉合成酶催化ADPG与淀粉引物（葡聚糖）反应，将葡萄糖分子转移到淀粉引物上并生成ADP，再通过丙酮酸激酶、己糖激酶和6-磷酸葡萄糖脱氢酶依次催化NADP+还原为NADPH，NADPH在340 nm处具有特征吸收峰，通过吸光值的变化即可表征结合态淀粉合成酶活性。
