检测原理
苹果酸脱氢酶能够催化苹果酸和NAD生成草酰乙酸和NADH,柠檬酸合酶可催化乙酰辅酶A和草酰乙酸生成柠檬酸和辅酶A,通过苹果酸脱氢酶和柠檬酸合酶的偶联反应体系,乙酰辅酶A含量和NADH的生成速率成正比,NADH在340 nm处具有特征吸收峰,通过吸光值的变化速率即可定量检测乙酰辅酶A的含量。

①准确在20 s和80 s处完成吸光值的测定,以确保实验结果的准确性和重复性;
