碱性蛋白酶能够在碱性条件下，催化酪蛋白水解产生酪氨酸，酪氨酸在碱性条件下能够还原磷钼酸化合物生成钨蓝，产物在680 nm处具有特征吸收峰，通过吸光值变化即可表征碱性蛋白酶的活性。
