谷胱甘肽过氧化物酶可催化H2O2氧化GSH生成GSSG，GSH和5,5’-二硫代-双-（2-硝基苯甲酸）反应生成黄色的2-硝基-5-巯基苯甲酸，产物在412 nm处有特征吸收峰，通过吸光值下降速率即可表征谷胱甘肽过氧化物酶活性。①若测定吸光值大于1.0，建议将粗酶液使用提取液适当稀释后再进行测定，计算时相应修改；
②显色完成后应立即测定吸光值，以免检测时间过长影响显色造成结果误差。
